چکیده

A line of C3H mouse embryo cells (C3H/10T1/2 clone 8) sensitive to postconfluence inhibition of cell division was used for a variety of quantitative and qualitative studies of chemical oncogenesis in culture. The polycyclic hydro carbons (PH), 3-methylcholanthrene, dibenz[a,h]anthracene, and 7,12-dimethylbenz[a]anthracene, caused varying degrees of cytotoxicity and produced morphologically and malignantly transformed foci in these cells with a dosedependent frequency, while yV-methyl-/V'-nitro-./V-nitroso-

Quantitative Studies of Radiation Transformation with the A31 -11 Mouse BALB/3T3 Cell Line1

The initial studies of X-ray transformation in vitro were carried out with primary hamster embryo cell cultures (4, 6). Recently, however, interest has centered on the use of established mouse embryo fibroblast cell lines which are highly sensitive to the postconfluence inhibition of cell division (7, 17, 20, 21 , 25). Such cell lines offer several advantages over primary cultures. Being establ...

O-18: Epigenetic Modification of Cloned Embryo Development; State of ART

Background: At the outset of the somatic cell nuclear transfer (SCNT) process, the chromatin structure of the somatic cell which governs its state of differentiation undergoes dramatic changes, called reprogramming, and is compelled back to the embryonic stage. However, the overall epigenetic makeup of the resultant cloned embryos has been acknowledged far different from the fertilized embryos....

Enhancement of X-ray transformation by 12-O-tetradecanoyl-phorbol-13-acetate in a cloned line of C3H mouse embryo cells.

A phorbol ester promoting agent, 12-O-tetradecanoyl-phorbol-13-acetate, enhances X-ray transformation in vitro in a two-stage fashion similar to that shown previously for ultraviolet radiation and chemical carcinogens. In studies with a mouse embryo-derived cell line (C3H/1OT 1/2 clone 8), there were clear interactive effects between X-radiation and 12-O-tetradecanoylphorbol-13-acetate. These w...

A Qualitative and Quantitative Assay for Cells Lacking Postconfluence Inhibition of Cell Division: Characterization of This Phenotype in Carcinogen-treated Syrian Hamster Embryo Cells in Culture1

We have developed a qualitative and quantitative assay system for detecting cells lacking postconfluence inhibition of cell division (contact insensitivity, CS~) in golden Syrian ham ster embryo cells in culture by measuring the number of cells able to form colonies on a lethally irradiated, confluent monolayer of a contact-sensitive established cell line. A subpopulation in normal low-passage ...
